Molecular Genetics of Cannabis Abuse

  • Agrawal et al Drug Alcohol Depend. 2008 Jan 11;93(1-2):12-20  report a maximum LOD of 1.9 is observed in the COGA sample at 95 cM on chromosome 14 for cannabis dependence.

  • Agrawal et al Drug Alcohol Depend. 2008 Dec 1;98(3):185-90 reports suggestive evidence for linkage on chromosome 18 (LOD 2.14 for frequency of  cannabis use, LOD 1.97 for cannabis initiation, at 90-97 cM) and also on chromosome 19 (LOD 1.92 for early-onset at 17 cM of cannabis use) in an Australian twin sample.

  • Agrawal et al Arch Gen Psychiatry. 2008 Jun;65(6):713-21 reports that genes such as ELTD1 on chromosome 1, in addition to genes on chromosomes 4 (eg, GABRA2) and 6 (eg, CNR1), may be associated with the genetic risk for cannabis use disorders as measured using a novel factor score.

  • SNPs in the CNR1 (rs2023239) and FAAH (rs324420) genes are associated with cannabis craving and withdrawal (Haughey et al. Addiction. 2008 Oct;103(10):1678-86)
